rum, using two techniques. One lot of fragments was implanted into the blastocoel ; the second lot was cultured in vitro in association with undetermined epiblast excised from the ventral half of the blastocoel roof. All samples were cultured up to 3 days in Barth’s solution, optimized for the amphibian embryo and not permitting isolated mouse egg cylinder fragments to survive. Small grafts, completely surrounded by Bufo cells, had an apparently normal aspect and frequent mitotic figures. Large grafts, attached to the host embryo, were in close contact with the endoblast and mesoblast cells, but the ectoblast layer showed no ten-

Histogenetic and neoplastic potential of different regions of the mouse embryonic egg cylinder.

The histogenetic and neoplastic potentials of defined regions of the 8th day mouse embryonic egg cylinder were examined following ectopic transfer to beneath the testis capsule. No differences in histogenetic potential were detected between anterior and posterior slices of the embryo, either when composed of all three germ layers or of embryonic ectoderm alone. Small anterior and distal fragmen...

Metabolic co-operation between embryonic and embryonal carcinoma cells of the mouse.

Mouse embryonal carcinoma (EC) cells form permeable junctions at their homotypic cell-to-cell contacts which permit intercellular exchange of metabolites (metabolic co-operation). Hooper & Slack (1977) showed how this exchange could be detected by autoradiography as the transfer of [3H]nucleotides between PC13 (a pluripotential EC line) and PC13TG8 (a variant of PC13 which is deficient in hypox...

Nutritional endoderm in a direct developing frog: a potential parallel to the evolution of the amniote egg.

The egg of the direct-developing frog, Eleutherodactylus coqui, has 20 x the volume as that of the model amphibian, Xenopus laevis. Increased egg size led to the origin of nutritional endoderm, a novel cell type that provides nutrition but does not differentiate into digestive tract tissues. As the E. coqui endoderm develops, a distinct boundary exists between differentiating intestinal cells a...

Regionalisation of the mouse embryonic ectoderm: allocation of prospective ectodermal tissues during gastrulation.

The regionalisation of cell fate in the embryonic ectoderm was studied by analyzing the distribution of graft-derived cells in the chimaeric embryo following grafting of wheat germ agglutinin--gold-labelled cells and culturing primitive-streak-stage mouse embryos. Embryonic ectoderm in the anterior region of the egg cylinder contributes to the neuroectoderm of the prosencephalon and mesencephal...

Failure of egg cylinder elongation and mesoderm induction in mouse embryos lacking the tumor suppressor smad2.

smad genes constitute a family of nine members whose products serve as intracellular mediators of transforming growth factor beta signals. SMAD2, which is a tumor suppressor involved in colorectal and lung cancer, has been shown to induce dorsal mesoderm in Xenopus laevis in response to transforming growth factor beta and activins. The smad2 gene is expressed ubiquitously during murine embryoge...
